RESUMO

This study was conducted to evaluate the effects of treatment with strontium ranelate (SR) on the repair of bone defects and molecular components of bones in femurs. Adult female rats (n=27) were subjected to ovariectomy (OVX) or Sham surgery. Thirty days after surgery, a defect was made in the femur and the animals were then divided into three groups: OVX, SHAM and OVX+SR. Euthanasia was performed four weeks after the bone defect surgery. Repair in bone defect was assessed by computed microtomography (µCT) and chemical composition of cortical bone was analyzed by Fourier transform infrared (FTIR) spectroscopy and energy dispersive X-ray spectroscopy (EDS). The trabecular thickness (Tb.Th) of the newly formed bone in the OVX+SR group was significantly higher than that for the OVX group. The collagen maturity in the OVX+SR group was smaller than in the other two groups. In this group, a significant increase in the amount of strontium (Sr) and a decrease in the amount of calcium (Ca) embedded to bone tissue were also observed. Systemic treatment with SR improved microarchitecture of the newly formed bone inside the defect, but decreased cross-linking of mature collagen in cortical bone.

Resumo Este estudo foi conduzido para avaliar os efeitos do tratamento com ranelato de estrôncio (RE) na reparação de defeitos ósseos e componentes moleculares de ossos nos fêmures. Ratas adultas (n = 27) foram submetidas a ovariectomia (OVX) ou cirurgia Sham. Trinta dias após a cirurgia, um defeito foi feito no fêmur e os animais foram então divididos em três grupos: OVX, SHAM e OVX+RE. A eutanásia foi realizada quatro semanas após a cirurgia de preparo do defeito ósseo. A reparação do defeito ósseo foi avaliada por microtomografia computorizada (μCT) e a composição química do osso cortical foi analisada por espectroscopia de infravermelho de transformada de Fourier (FTIR) e espectroscopia por energia dispersiva de raios X (EDS). A espessura do osso trabecular (Tb.Th) recém formado no grupo OVX+SR foi significativamente maior que a do grupo OVX. A maturidade do colágeno no grupo OVX+SR foi menor do que nos outros dois grupos. Neste grupo, observou-se também um aumento significativo na quantidade de estrôncio (Sr) e uma diminuição na quantidade de cálcio (Ca) no tecido ósseo. O tratamento sistêmico com RE melhorou a microarquitetura do osso recém formado dentro do defeito, mas diminuiu a reticulação do colágeno maduro no osso cortical.

O ranelato de estrôncio (RS) diminui a reabsorção óssea e ao mesmo tempo age como um agente anabólico deste tecido. Este estudo foi realizado para avaliar os efeitos da ovariectomia e do tratamento com RS na reparação de defeitos ósseos em fêmures e nos componentes moleculares do osso em ratas. Vinte e sete ratas adultas foram submetidas a ovariectomia ou cirurgia Sham e, após trinta dias, defeitos ósseos em fêmures foram confeccionados e os animais divididos em três grupos: ovariectomizadas (OVZ), cirurgia Sham (SHAM) e ovariectomizadas + tratamento com 625 mg/kg/dia de RS (RS). A eutanásia foi realizada quatro semanas após a cirurgia do defeito ósseo. A reparação do defeito ósseo foi analisada por microtomografia computadorizada (?CT) e a composição química do tecido ósseo foi verificada por espectroscopia de infravermelho com transformada de Fourier (FTIR) e espectroscopia de energia dispersiva de raios-X (EDS). O grupo SHAM apresentou em média volume ósseo (BV) superior ao grupo OVZ (p=0,014) e o volume ósseo relativo (BV/TV) foi cerca de 8% superior ao grupo OVZ. A comparação entre SHAM e RS apresentou valores limítrofes para essas variáveis. A espessura trabecular (Tb.Th) no grupo RS foi significativamente maior que no grupo OVZ cerca de 1,8% (p = 0,049) e não diferiu em média do grupo SHAM. Não houve diferenças significativas nas relações que avaliam compostos inorgânicos sobre orgânicos nos grupos...


Strontium ranelate (SR) decreases bone resorption and at the same time acts as an anabolic agent in this tissue. This study was conducted to evaluate the effects of ovariectomy and treatment with SR on the repair of bone defects and molecular components of bones in femurs. Adult female rats (n=27) were subjected to ovariectomy or Sham surgery. Thirty days after surgery, a defect was made in the femur and the animals were divided into three groups: ovariectomy (OVZ), Sham surgery (SHAM) and ovariectomy plus treatment with SR, 625 mg / kg / day (SR). Euthanasia was performed four weeks after surgery to cause the bone defect. Repair in bone defect was assessed by computed microtomography (?CT) and the chemical composition of bone tissue was analyzed by Fourier transform infrared (FTIR) spectroscopy and energy dispersive X-ray spectroscopy (EDS). The SHAM group showed mean values for bone volume (BV) higher than those of the OVZ group (p = 0.014) and the relative bone volume (BV / TV) was about 8% higher than that of the OVZ group. Comparison between SHAM and SR showed borderline values for these variables. In the SR group, the average value for trabecular thickness (Tb.Th) was significantly higher (~1.8%) than that in the OVZ (p = 0.049) group and did not differ from that of the SHAM group. Significant difference was not observed in the relation between inorganic and organic compounds in the three groups. The ratio of the areas of the bands in 1057 and 1023 cm-1 for the calculation of IC was 1,024 in the SHAM group, 1,015 in the RS group and 1,108 in OVZ group. In the study of the amide III band, a change in the proportion of secondary structure of matrix proteins was observed in ovariectomized animals, possibly a decrease in of ?-helical and random coil (RC) and an increase in ?-sheet structures...

The aim of the present study was to investigate the relationship between degenerative bone changes of the head of the mandible and the presence of joint effusion (JE). This study was based on sagittal magnetic resonance imaging (MRI) reports of 148 temporomandibular joints (TMJs) of 74 patients complaining of pain and/or dysfunction in the TMJ area. The mandible heads were surveyed for osteoarthritis characteristics, which were classified as osteophytosis, sclerosis or erosion. The presence of JE was checked whenever high signal intensity was observed in the articular space. The results evidenced the presence of bone changes in 30% of the sample. Osteophytes and erosions were the changes most commonly observed. JE was reported in 10% of TMJs. The results from the statistical tests revealed that bone changes in the head of the mandible are associated with the presence of JE.

Esta pesquisa avaliou o efeito do uso local do residronato de sódio, em diferentes concentrações molares, no processo de reparação de defeitos ósseos em fêmures de ratas. Foi confeccionado no fêmur de 60 ratas um defeito ósseo medindo 2,5 mm de diâmetro. Estes animais foram divididos em grupos: controle (C), amido (Am), residronato 0,25 mol (Res 0,25), residronato 0,5 mol (Res 0,50), residronato 0,75 mol (Res 0,75) e residronato 1 mol (Res 1), de acordo com o material de preenchimento utilizado. Nos animais do grupo controle o defeito ficou preenchido apenas pelo coágulo. Os animais foram eutanasiados aos sete e 21 dias,e o fêmur foi removido, fixado e descalcificado, para a confecção delâminas histológicas. Foram realizadas análises histológica e histomorfométrica, e os dados obtidos foram submetidos à análise estatística ANOVA. Histologicamente, as principais diferenças ocorreram após 21 dias. Os grupos C e Am apresentaram fechamento em extensão do defeito ósseo em todos os espécimes e a maioria dos animais dos grupos tratados com residronato em diferentes concentrações molares não exibiu neoformação óssea na região central do defeito, permanecendo este preenchido por tecido conjuntivo. Nos períodos de sete e 21 dias não houve diferença estatística significante entre os grupos C e Am em relação a neoformação óssea. Estatisticamente, não houve diferença entre os grupos Res 0,25; Res 0,50; Res 0,75 e Res 1 no período de sete dias. Já no período de 21 dias o grupo Res 0,25 apresentou a maior média, mas não diferiu estatisticamente do grupo Res0,75. Concluiu-se que o residronato de sódio em todas as concentrações molares, prejudicou a reparação óssea nesse modelo experimental. No entanto, foi observada uma neoformação óssea extra-cortical subperiosteal nos grupos que receberam residronato


This work evaluated the action of sodium residronate, in different molars concentrations, in the repair of bone defects in femurs of rats. A bone defect measuring 2,5 mm in diameter was made in the femur of 60 rats.These animals were divided in groups: control (C), starch (Am),residronate 0,25 mol (Res 0,25), residronate 0,50 mol (Res 0,50),residronate 0,75 mol (Res 0,75), residronate 1 mol (Res 1), in accordance with the material of fill used. In the animals of the control group, the defect was just filled by the clot originating from the defect. The animals were euthanized at seven and 21 days, when the femur was removed, fixed and decalcified, to make histologicals lamina e. Histological and histomorphometric analyses were performed and the results obtained were submitted to statistical analysis A NOVA. Histologically, the principal differences occurred after 21 days. The groups C and Am, showed an extension closure of bone defect in every specimen and most animals in groups treated with residronate in different molar concentrations did not show bone neoformation in the central region of the bone defects, which remained filled with connective tissue. After seven and 21 days, there was no significant statistical difference among groups C and Am in relation tobone neoformation. Statistically, there were no differences among thegroups Res 0,25; Res 0,50; Res 0,75 e Res 1 in the period of seven days.In the period of 21 days, the group Res 0,25 had the highest average, butdid not differ statistically from the Res 0,75. It was concluded that the sodium residronate in all the molar concentrations, harmed the bone repairin this experimental model. Ho wever, there was an extracortical subperiosteal bone neoformation in groups receiving residronate
